Puedes postear tu web.config, estoy casi casi seguro que ahí está el problema.! EDIT: Sí, estuve googleando un poco, porque ese error nunca lo había visto, y bueno, encontré lo siguiente en un blog: If you're trying to use ASP.NET AJAX features (like Script Manager or Update Panel) in your ASP.NET 3.x website and getting "ASP.NET AJAX client-side framework failed to load" error, chances are your Web.config file is missing required sections. Fastest way to correct this is to create a new ASP.NET website in Visual Studio (you can delete it afterwards) and copy missing sections from the new web.config to the one in your web site. Haz eso a ver que tal. Saludos!

Ahhh claro, no se por qué no lo ví.
Fíjate, este es el método que quieres llamar: Button2_Click(ByVal sender As System.Object, ByVal e As System.EventArgs)
Tiene dos parámetros o argumentos. Tienes que pasarle algo ahí.
puedes hacer algo así:
Button2_Click(Me, EventArgs.Empty)
Y me dices como te fue.

Claro, porque si lees la especificación, luego de que carga todo el contenido del html, debes ver todos los componentes que deben ser descargados, es decir, que no vienen junto con el texto del HTML.
Entonces, tendrías que analizar las peticiones para regresar no solo el HTML sino también imágenes, objects, video (en caso de html5), etc.
